Sequencing of a fragment of the S-Adenosine methionine decarboxylase (SAMDC) gene in pistachio (Pistaciavera) cv. Badami-Zarand

One of the most important problems in pistachio cultivation industry in Iran is the reduced yield of pistachio caused by increased soil and water salinity. Polyamines, as a natural phytocompound, play a major role in response to salinity. In the current research, identification of the sequence of one of the genes involved in biosynthesis of polyamines, i.e. S-Adenosine methionine decarboxylase (SAMDC),was investigated in the pistachio (Pistaciavera) cv. Badami-Zarandby using degenerate and designed primers on the basis of close plants. Sequence of a fragmentof the gene of 462 nucleotidesize was identified for the first time in pistachio. Therefore, by identification of this sequence, the first step towards complete sequencing of the gene has been taken. Also, the results of nucleotide and aminoacid sequence alignment of this fragment showed high similarity with this gene in other plants and the extent of this similarity was higher in phylogeneticallyimminentplants.
