An efficient micropropagation protocol based on multiple shoot induction has standardized in Crotalaria retusa. High frequency of multiple shoots has observed on MS medium supplemented with BAP (13.31 μM). On this medium, 100% cultures responded with an average number of 11.6 shoots per explants. However, the average shoot length was limited to 3.2 cm. The addition of 2.15 μM NAA along with 13.31 μM BAP gave to an average number of 12.4 shoots with an average shoot length of 6.2 cm. Luxuriantly growing callus was obtained on MS medium supplemented with BAP (2.21 μM) and 2,4-D (13.57 μM). Optimum callus regeneration obtained on MS medium supplemented with 13.31 μM BAP and 2.15 μM NAA. On this medium, 96.4 % cultures responded with an average number of 12.6 shoots per culture. The shoots obtained via multiple shoot induction and organogenesis was rooted on half-strength MS medium supplemented with IBA (7.38 μM). The rooted shoots were successfully transplanted to soil with 90% success. The standardized protocol would help to conserve this medicinal plant.
