The enzyme L-asparaginase has been thoroughly researched by many researchers worldwide because of its
immensely useful medical applications. It has proved to be an efficient chemotherapeutic agent especially in the
treatment of Acute Lymphoblastic Leukemia [ALL] and Lymphosarcomas. The present paper discuss the studies
carried out for the production of this enzyme from E. coli K-12, followed by the optimization of the LB media
specified for the optimal production of the enzyme. On the basis of results obtained from media optimization studies,
a modified production media has been developed for optimal L-asparaginase production. The enzyme activity
(1.01±0.8 IU/ml) markedly increased in the modified production medium (3.82±0.1 IU/ml).
